Method Introduction
Ultra-high-performance size-exclusion chromatography (UP-SEC) is a variation of the more established HP-SEC technique.
In principle, a UP-SEC system works like an HP-SEC instrument but operates under much-increased system pressures of up to 1,000 bar. This reduces analysis time, requires lower sample and mobile phase volumes and delivers better resolution.
The high pressures inside a UP-SEC system demand advanced column set-up with specially designed beads. Thus, this technique is less versatile than HP-SEC due to the limited availability of different columns.
Applications
UP-SEC can be employed in all stages of biopharmaceutical research and development and for lot release testing. If increased sample throughput or better resolution is not required, HP-SEC, a more established technology, is preferred.
