Colorimetry
Method Introduction
Colorimetry is a crucial analytical technique employed to measure the degree of coloration in a solution by evaluating its color intensity. This spectrophotometric method is based on the European Pharmacopoeia, where transmittance values are obtained at wavelengths throughout the visible spectrum (380 nm – 720 nm) and compared to specified color reference solutions. Colorimetry is essential for assessing the quality and stability of biopharmaceutical products, as color changes can indicate chemical, physical or biological instabilities including but not limited to degradation or contamination.
Applications
Colorimetry is widely used in various stages of biopharmaceutical development and manufacturing. It is particularly valuable for:
- Monitoring the stability of formulations by detecting color changes over time.
- Performing quality control and release testing of drug products.
- Evaluating the presence of impurities and contaminants in formulations.
- Supporting forced degradation studies by assessing color changes under stress conditions.
