Mass Photometry
Method Introduction
Mass photometry (MP) is a light-scattering-based technique that enables the accurate mass measurement of individual biomolecules in solution, in their native state.
In mass photometry, the attachment of a molecule to a cover glass causes a local change in reflectivity at the glass-water interface, which is acquired as a local change in contrast in the microscopy image. The determined contrast values are converted to molecular mass based on a calibration with proteins of known molecular weight. The TwoMP instrument can detect molecules in the molecular mass range from 40 kDa to 5 MDa. MP is based on a single particle detection principle. A mass distribution plot allows to differentiate particle populations by their molecular weight and provides information about the relative abundance of each particle population

Applications
MP enables a rapid empty vs. full AAV capsid characterization, as its mass resolution allows differentiation between empty and genome filled capsids with minimal sample amount. MP can be used to measure molecular masses of proteins and nucleic acids, to study the heterogeneity of large protein complexes and to characterize protein-protein interactions, such as antibody-antigen affinities.
