Total Holographic Characterization
Method Introduction
Total holographic characterization (THC) is a quantitative technique for the analysis of subvisible particles enabling to classify particles based on their refractive index.
In THC, particles flow through laser light in a microfluidic channel and the interference pattern of scattered laser light is recorded for each particle. Comparison of the interference pattern (measured hologram) to the scattering theory by Lorenz-Mie yields particle size, refractive index and symmetry of each single particle. A distribution plot allows to differentiate particle populations by size and refractive index in a size range of 0.5 µm to 10 µm. Sample concentration is determined by counting of particles per analyzed volume.

Applications
THC can be used to differentiate particles by their refractive index, such as protein particles from silicone oil droplets. Compared to the orthogonal technique of resonant mass measurement (RMM), THC includes a larger size range up to 10 µm.
